pEF1a-BirA-VP16

This part is used to constitutionally express BirA-VP16 in TIL cell(The tumor infiltrates lymphocytes ) , which will help to generate biotin induction system. Promoter EF1a is a strong promoter and it will constitutionally drive BirA-VP16 expression. Through lentiviral transfection, this part will insert genome DNA of TIL cell. When biotin is present, BirA-VP16 will bind biotin and change its conformation, and then recognize operon of birA. With the help of VP16,a transcriptional activator from herpes simplex virus, BirA-vp16 will activate miR-135b-3p expression in TIL Cell.
